C.elegans RNAi - Library Plates and individual Bacterial
Strains
The bacterial strains in the 384-well library plates have been grown in
LB broth containing 8% glycerol and 25µg/ml carbenicillin. The plates
have been shipped to you on dry ice and should be frozen at -70°C
on receipt.
Individual bacterial strains have been streaked onto LB
agar containing 25µg/ml carbenicillin. The agar slopes should be
stored at 4°C on receipt.
If you encounter problems growing clones, please streak
for single colonies onto LB agar containing 50µg/ml Ampicillin and
15µg/ml Tetracycline.
Please see the accompanying Ahringer Lab protocol for use
of this library.
There is also a "
C.elegans Finder" tool available which allows you to find
the primer sequences for any given GenePair Name in addition to the location
of that bacterial strain in Geneservice Ltd. 384-well plates. The complete C.elegans
RNAi database can be downloaded by right
clicking here.
Every effort has been made to ensure that the plates that
you receive are as close as possible to the published database. However
in some plates, there may be empty wells due to a clone not growing -
please inform us if this happens and we will try to replace this clone
if possible. In the last plate of each set, due to plates having been
condensed from 96 well into 384 well plates- there may be only eg only
1 x 96 well plate resulting in 75% of the plate being empty. If any growth
is observed in a well thatshould be empty, please ignore this, it may
be due to very low level cross contamination.
For the re-arrayed subsets please consult the following datasheets
- Chromatin
- Phosphatase
- Transcription factors

On your Despatch Note a Bacterial Strain is called a "CLONE".
