In many cases, the individual chromosome was flow-sorted
from a rodent/hybrid cell line containing the single human
chromosome of interest (Trask
et al 1991 Som, Cell Mol. Gen. 17 117-136). The flow sorting
was done using a high-speed modular flow sorter designed and
built by LLNL engineers. Details on the methods used to produce
these libraries have been published: (Gingrich
et al 1996 Construction and Characterization of Human Chromosome-2-Specific
Cosmid, Fosmid and PAC Clone Libraries. Genomics 32 65-74).

IMPORTANT INFORMATION ON REQUESTING CLONES FROM LLNL
COSMID LIBRARIES
CLONES FROM ALL CHROMOSOME LIBRARIES (other than 19)
The LLNL libraries exist both in a 96-well plate and a 384-well
plate format, and it is essential that there is no confusion
in the databases (e.g. GDB) between clones identified from
these different formats.
The LLNL libraries now at Geneservice were obtained
in a 384-well format in June 1996. Prior to that time they
only existed in the 96-well format. Consequently all database
entries and publications are in the 96-well format.
It has been decided that this will be continued so that
there is no ambiguity in the identity of LLNL clones.
Therefore please regard the coordinates that you give to
your clones when requested as a result of screening filters
provided by Geneservice as CODE NAMES ONLY.
When we receive your request we will translate your code
names into the correct CLONE NAMES (i.e. the 96 well coordinates).
It is these CLONE NAMES that should be used in all publications
and database entries.
We can only be sure that we are providing you with the correct
clone if we know how you identified the clone ID. For this
reason, it is essential that you complete the section on our
request forms asking you for the source of your clone ID.
If
users wish to convert from 96 to 384 well coordinates (and
vice versa) they can use this utility.
CLONES FROM CHROMOSOME 19 LIBRARIES
Clones from the three Chromosome 19 libraries, F, R and
W, should continue to be referred to in publications or database
entries by either the LLNL internal ID or the GenBank Clone
Name. For a full description of the nomenclature see click
here.
Please regard the coordinates that you give to your clones
when requested as a result of screening filters provided by
Geneservice as CODE NAMES ONLY.
When we receive your request we will translate your code
names into the correct CLONE NAMES (i.e. the 96 well coordinates).
It is these CLONE NAMES that should be used in all publications
and database entries. On your despatch note the CLONE NAME
we will use will be in the GenBank format (e.g. LLNLR-310G1)
but when submitting data it would be helpful to include the
LLNL internal ID (e.g. R34737) in addition.
We provide a tool for correlating all the IDs for a given
Chromosome 19 clone at Chromosome19_Finder
