Drosophila cosmid clones - an explanation
of your Dispatch Note
All publications and database entries for this Drosophila cosmid
library must be in the 96-well format. Below is an example of a Despatch
Note printout indicating the correct ID to use:
Drosophila cosmid clones
(in E.coli)
Clones are streaked onto LB agar containing kanamycin (25 µg/ml).
To use: re-streak the clone onto the same agar type, in such a way as
to obtain single colonies. Incubate at 37°C overnight.
Single colonies are then ready for DNA isolation, using standard protocols,
if required.
Clones can be grown in LB broth + kanamycin, and glycerol
added after growth to a concentration of 25% for long term storage at
-70°C.
The vector used is Lorist 6 (Gibson. TJ. et al [1987]
Gene 53: 283-286).
