Mouse cDNA NIA 7.4K and 15K Clones
Clones are supplied streaked onto LB agar + ampicillin (100µg/ml).
Clones on agar should be stored at 4°C on receipt.
As with all clones it is recommended that you re-streak to obtain several
single colonies to test. Clones that test positive can be stored long-term
as glycerol stocks at -70°C. After growth in LB broth + antibiotic,
glycerol is added to a final concentration of 25%.
This clone set was constructed in Minoru Ko's laboratory at the Laboratory
of Genetics at the National Institute on Aging (NIA), National Institutes
of Health (NIH) Baltimore, USA., using tissue derived from embryonic and
fetal cells from C57BL/6J mice. The vector used is pSPORT1 (see map below)
[Invitrogen] and the host strain is E.coli DH10B. The cDNA inserts were
cloned into the vector using Not1 and Sal1 restriction sites. The following
Forward and Reverse primers can be used for cDNA insert amplification
or sequencing:
Ko forward primer: GTG TGG AAT TGT GAG CGG ATA ACA A (25mer)
Ko reverse primer: CCA GTC ACG ACG TTG TAA AAC GAC (24mer)
Clone names
There are 79 x 96 well plates in the 7.4k clone set, numbered H4001-H4079.
There are 159 x 96-well plates in the 15k clone set, numbered H3001 -
H3159. Please adhere to the following clone name format in any publication:
Clone H12 from plate H3145 should be referred to as H3145H12
